Furosemide (F)-induced nephrocalcinosis (NC) has been traditionally described in LBW prematures. A recent article (J Peds 1994;125:149) reported this complication in older infants with
congestive heart failure. To investigate the role of age on F-induced NC we studied 32 Sprague-Dawley male rats grouped by age and F therapy vs. control for 14 days. The rats were placed in
metabolic cages for measurement of urine output, food and H2O intake. At the end they were anesthetized and exsanguinated. Renal calcium deposition was graded using NC score (scale 0-4) and
by the calcium content(ugm/gm dry tissue) in the contralateral kidney. Table By both indices controls had no NC. All treated groups developed NC with no difference in its magnitude among
themselves. Despite similar initial weights and food intake/100 gm dry wt, treated animals grew less and had higher fluid intake and urine output than their age-matched controls. The more
mature F-treated groups (F and H) had hypokalemia, hypochloremia and increased urinary losses of K and Mg. There were no differences between treated and control groups with regards to Cr
clearance, serum Cr, P and iCa nor in urine Ca, P and Mg excretions/100 gm dry wt. We conclude that the development of NC following two weeks of F treatment is not influenced by age. AUTHOR
