Access through your institution Buy or subscribe Colorectal cancer stem cells (CSCs) can self-renew and contribute to colorectal tumorigenesis and metastasis. New research has revealed that
5-hydroxytryptamine (5-HT) produced by enteric neurons can modulate CSC self-renewal in mice. 5-HT receptors (HTR1B, HTR1D and HTR1F) were highly expressed in colorectal CSCs and engaged
with 5-HT to activate Wnt–β-catenin signalling. Moreover, a tumour-associated microbial metabolite (isovalerate) promoted 5-HT production. Crucially, blockade of 5-HT signalling in a mouse
model of CRC inhibited CSC self-renewal but also suppressed tumour progression and metastasis (from intestine to liver). In human colorectal tumours, 5-HT levels and Wnt signalling
